The Monash Antibody Discovery Platform (MADP) provides services for antibody production, assay development, robotics, high throughput screening and antibody characterisation, including monoclonal antibody sequencing. We use state-of-the-art equipment to support researchers not only from Monash University but elsewhere in Victoria, Australia and world-wide.
The primary focus in the platform is generating high-affinity monoclonal antibodies through advanced discovery techniques and antibody engineering. However, we are keen to help customers with their antibody or laboratory automation challenges. We are happy to develop new antibody screens and encourage our customers to bring their requests to us.
The staff from MADP work with our customers at every step to ensure the needs of each individual project are met and we are proud to follow a Quality Management System that is certified to ISO9001.

Mycoplasma testing |
We can test all cell lines for the presence of mycoplasma using a commercial kit that can detect the Mycoplasma and Acholeplasma species that most commonly contaminate cell culture. These include the six species that account for 95% of all contaminations: M. orale, M. hyorhinis, M. arginini, M. fermentans, M. hominis, and A. laidlawii. We confirm all positive results with an additional PCR reaction if required. We can also treat cells to remove mycoplasma contaminations if a culture tests positive. |

Cell-line banking (freeze 20 vials per cell-line) |
For cell line banking we freeze 2 x 10 vials of cells and store across 2 separate tanks at Monash University. If required we can send some, or all, of the frozen cell vials to an address as specified by the customer for storage off-site. |

Monoclonal antibody sequencing (DNA) |
We can isotype and sequence mouse or rat antibodies from hybridoma cell lines generated at the facility as well as cell lines provided by customers. Knowing the sequence of your antibody is essential for mammalian expression, humanisation or for IP protection. The service includes the cloning and sequencing of the light and heavy chain variable regions and typically takes 2-4 weeks. This monoclonal antibody sequencing service requires a frozen cell vial or growing hybridoma culture* for the sequencing of the heavy-chain variable region, (VH), and the light-chain variable region, (VL), of the monoclonal antibody. Data will be analysed using the IMGT/V-Quest program (The International Immunogenetics Information System) and VH and VL sequences are compared to unrearranged germline mouse antibody genes. A project report will be provided at the end of the sequencing in which the VH and VL protein sequences are annotated to highlight framework regions (FRs), and complementary determining regions (CDRs).

Monoclonal antibody sequencing (Protein) |
We can determine the amino acid sequence of an unknown antibody using mass spectrometry from bottom-up proteomics techniques with multiple enzyme digests. Antibody protein sequencing does not require the hybridoma cell line or the DNA sequence and we can work with as little as 20µg material. This service requires 20ug material at ~1mg/mL The antibody sample is run on a gel to identify two bands (approx. 20kda and above 50kDa to meet normal expectations for an antibody sample). The sample is then digested with three different enzymes analysed by LCMS and the resulting data processed with the PeaksAB software and manually refined. The final sequences will be provided in a report. |

SPR determination of mAb pair or binding kinetics |
Surface Plasmon Resonance (SPR) is a biosensor technology enabling label-free and real- time measurement of protein-protein interactions. In the capture assay, the ligand of interest binds to the specific antibody that is immobilized on a chip. The kinetics of binding can then be measured. The principle uses of SPR analysis are to identify complimentary binding pairs of antibodies with recombinant antigen or to determine binding kinetics and affinity of antibodies with the parent antigen. Our SPR projects are run in collaboration with the Monash Fragment Platform
